Residual Moisture and Karl Fischer Measurement

A solid that appears dry can contain water invisible to the eye. Karl Fischer quantifies it through a chemical determination. The result describes a sample and a method; it does not automatically establish stability or explain where all that water was distributed.

What the determination represents

USP chapter 921 distinguishes procedures for water determination. Karl Fischer includes volumetric and coulometric variants: they share an analytical reaction, but do not obtain quantity through the same way of supplying and measuring reagent.

The word moisture on a certificate therefore needs an associated method. It should not be assumed automatically equivalent to mass loss during drying, where the observed change may answer a different analytical question.

The sample can also contribute to error

The USP text highlights chemical compatibility, possible side reactions and contributions of environmental moisture to the system. These factors matter especially when sample water is small relative to other contributions.

To interpret a result, determine whether the material was examined directly or through a preparation, and which controls support the measurement. The instrument's name alone does not establish that every signal corresponds to water originally present in the solid.

Water content and sorption behavior differ

A 1997 investigation compared water-coverage estimates in freeze-dried proteins using molecular composition, Karl Fischer titrations and gravimetric sorption analysis. Results agreed for the studied proteins.

Crystalline mannitol, however, did not support the interpretation used for other materials. The example shows that a sorption model has assumptions about physical state. Measuring water and explaining how it associates with a formulation are related but not interchangeable tasks.

An indirect method needs a reference

In 2021, noninvasive spectroscopic techniques were compared with Karl Fischer in a freeze-dried enzyme-and-sucrose formulation. Agreement depended on moisture range and preparation of the comparison samples.

The spectroscopic models had greater difficulty at the low-moisture end studied. That observation belongs to those models and materials; it does not show universal inferiority of spectroscopy. It does, however, prevent transferring a calibration between formulations without checking performance.

What the certificate can conclude

A complete result preserves sample identity and state, method and basis of expression. When compared with an indirect technique, the range over which agreement was demonstrated also matters.

Water percentage is a characterization attribute. Linking it to stability requires additional observations of the formulation over time. An isolated figure cannot establish storage duration, molecular integrity or behavior of every preparation with the same ingredient.
